It specifically targets the voltage-gated potassium channel hERG (KCNH2) with a half-maximal inhibitory concentration (IC50) of 1 μM. Beyond its primary function, Amiodarone Hydrochloride exhibits high affinity binding to the sigma-1 opioid receptor, 3-β-hydroxysteroid Δ8Δ7 isomerase, and C-8 sterol isomerase, with dissociation constant (Ki) values of 1, 25, and 62 nM, respectively. Furthermore, it inhibits human thyroid hormone receptors α and β, with IC50 values of 0.6 and 0.65 μM. It also operates as an inhibitor of cytochrome P450 (CYP) isoforms CYP2C8 and CYP3A4 at low micromolar concentrations in vitro, underscoring its multifaceted pharmacological capabilities.
