Application
Methyl-β-cyclodextrin is primarily utilized as a cholesterol-removing agent, effectively disrupting lipid rafts. Due to its high solubility and affinity for detergents commonly used in membrane protein chemistry, it is also employed to facilitate the removal of detergents from protein-lipid-detergent mixtures, thereby aiding in the formation of 2D membrane protein crystals. This compound, a randomly methylated form of the cyclic oligosaccharide β-cyclodextrin, consists of seven D-(+)-glucopyranose units with randomized hydrogen or methyl groups. Methyl-β-cyclodextrin enhances the aqueous solubility of various compounds and extracts cholesterol from lipid membranes. Additionally, at a concentration of 5 mM, it has been shown to reduce α-synuclein levels in membrane and detergent-insoluble fractions of B103 neuroblastoma cells, which are transfected with human α-synuclein. It also decreases α-synuclein levels in the brains of mice in a transgenic model of α-synucleinopathy.
